Design of the RNA Communication Circuit for Cell Mimics To engineer RNA-based communication between cell mimics, we chose an AND gate consisting of two small, synthetic RNA activators. (37) To activate protein production, the AND gate requires the presence of both RNA activators: A short RNA that activates transcription (STAR) (35) and a short trigger RNA that activates a toehold switch to allow translation.

Abstract On-demand biomanufacturing has the potential to improve healthcare and self-sufficiency during space missions. Cell-free transcription and translation reactions combined with DNA blueprints can produce promising therapeutics like bacteriophages and virus-like particles. However, how space conditions affect the synthesis and self-assembly of such complex multi-protein structures is unknown.
