Fludarabine combined with treosulfan or total body irradiation as a conditioning therapy before allogeneic hematopoietic stem cell transplantation in acute lymphoblastic leukemia. A retrospective matched-pair analysis from the ALWP of the EBMT
Abstract There are very limited data on treosulfan (Treo)-based conditioning before allogeneic hematopoietic stem cell transplantation (alloHSCT) in acute lymphoblastic leukemia. We aimed to compare fludarabine with Treo (FluTreo) or total body irradiation (FluTBI) in this setting. The final matched-pair analysis included ALL patients transplanted in complete remission after FluTreo (n = 153) or FluTBI (n = 431). The median patient age was 56.5 and 54.1 years, respectively.
Allogeneic hematopoietic cell transplantation for T-cell/histiocyte-rich large B-cell lymphoma: An EBMT lymphoma working party study
|
Abstract To the Editor T-cell/histiocyte-rich LBCL (THRLBCL) is an uncommon entity within the large B-cell lymphoma family, comprising less than 5% of all LBCL [1]. Compared to other LBCL, THRLBCL patients are younger, commonly male, present with higher International Prognostic Indexes (IPI), and advanced disease stages [2]. THRLBCL is named for its unique immunomodulating micro-milieu characterized by T-cell and histiocyte infiltration.

Anti-thymocyte globulin (ATG)- or alemtuzumab-based graft-versus-host disease prophylaxis in reduced-intensity conditioning allogeneic hematopoietic cell transplantation (HCT) for patients 40 years and older with acute lymphoblastic leukemia in first comp
|
Abstract For patients with high-risk acute lymphoblastic leukemia (ALL), allogeneic hematopoietic cell transplantation (HCT) remains standard of care. In the setting of an HLA-matched unrelated donor HCT, in vivo T-cell depletion (TCD) for prophylaxis of graft versus host disease (GVHD) relies on anti-thymocyte globulin (ATG) in Europe and alemtuzumab in the UK.

Reduced intensity conditioning with 8 Gy total body irradiation in adult patients with acute lymphoblastic leukemia - Bone Marrow Transplantation
Abstract Total body irradiation (TBI) plus chemotherapy is commonly applied prior to allogeneic hematopoietic stem cell transplantation (SCT) for patients with acute lymphoblastic leukemia (ALL).
Frequency and impact of somatic co-occurring mutations on post-transplant outcomes in acute myeloid leukemia: a multicenter registry analysis on behalf of the EBMT ALWP | Bone Marrow Transplantation
Abstract Acute myeloid leukemia (AML) includes genetically defined subsets. In allogeneic hematopoietic cell transplantation (allo-HCT), the frequency and prognosis of gene-gene interactions may differ from those of patients treated with chemotherapy alone. In this study, adult patients (N = 952) with AML allografted between 2015 and 2023, with available next generation sequencing (NGS) at diagnosis were included.
Genetic and epigenetic alterations at secondary resistance after continued decitabine-based treatment of acute myeloid leukemia in the randomized phase II DECIDER trial | Leukemia
Therapies containing the hypomethylating agents (HMA) decitabine (DEC) or azacitidine (AZA) are standard of care for patients with acute myeloid leukemia (AML) ineligible for intensive chemotherapy. However, initially responsive disease eventually develops secondary resistance [1,2,3,4].
Young unrelated donors confer a survival advantage for patients with myeloid malignancies compared to older siblings - Leukemia
Abstract Donor age is one factor to optimize allogeneic hematopoietic cell transplantation (alloHCT). Therefore, we investigated whether young unrelated donors (UD) provide a benefit for older patients with myeloid malignancies compared to HLA-identical sibling donors (MSD). We performed a retrospective registry study on patients ≥50 years who received a first alloHCT between 2010 and 2020.
Prognostic significance of cytogenetic risk score in patients with secondary acute myeloid leukemia undergoing allogeneic stem cell transplantation from HLA-matched unrelated donors: a study from the ALWP /EBMT | Bone Marrow Transplantation
Abstract The cytogenetic risk category retains a pivotal role in the prediction of prognosis in acute myeloid leukemia (AML) patients undergoing hematopoietic stem cell transplantation (HSCT), however, its impact on secondary AML (sAML) is less established. We assessed whether the ELN 2022 cytogenetic risk score predicts outcomes in sAML patients in remission undergoing HSCT from HLA-matched donors performed between 2010 and 2022.
Growing adoption of pharmacologic maintenance therapy after allogeneic hematopoietic cell transplantation in acute myeloid leukemia: a survey on behalf of the EBMT acute leukemia working party | Bone Marrow Transplantation
|
TO THE EDITOR: Disease recurrence in acute myeloid leukemia (AML) is the primary cause of failure in allogeneic hematopoietic cell transplantation (allo-HCT). The risk of relapse post-transplant is influenced by several factors, including the AML genomic profile, disease status at transplant, conditioning regimen, and measurable residual disease (MRD) presence before allo-HCT [1].

Allogeneic hematopoietic cell transplantation after infection with SARS-CoV-2 during the COVID-19 pandemic: a multicenter retrospective analysis - Bone Marrow Transplantation
To the Editor: At the onset of the COVID-19 pandemic, transplanters faced the unprecedented challenge of balancing the unknown risks of a SARS-CoV-2 infection in patients requiring allogeneic haematopoietic stem cell transplantation (alloHCT) against the benefits of this curative procedure.
The role of allogeneic stem cell transplantation in acute myeloid leukemia with translocation t(8;16)(p11;p13)
1 INTRODUCTION Translocation (8;16)(p11;p13) is a rare abnormality in acute myeloid leukemia (AML) with female preponderance, occurring more frequently in secondary AML (sAML), and especially in therapy-related (tAML) following exposure to topoisomerase-2 inhibitors.1, 2 Clinically, t(8;16) AML is associated with disseminated intravascular coagulation, high risk of bleeding, extramedullary involvement, and hemophagocytosis.1, 3 The translocation leads to the fusion of acetyltransferase KAT6A...
Tacrolimus versus cyclosporine a combined with post-transplantation cyclophosphamide for AML In first complete remission: a study from the acute leukemia working party (EBMT) | Bone Marrow Transplantation
Abstract Choice of calcineurin inhibitor may impact the outcome of patients undergoing T-cell replete hematopoietic cell transplantation (HCT) with post-transplant cyclophosphamide (PT-Cy) and mycophenolate mofetil (MMF) for prophylaxis of graft-versus-host disease (GVHD).
Remission induction versus immediate allogeneic haematopoietic stem cell transplantation for patients with relapsed or poor responsive acute myeloid leukaemia (ASAP): a randomised, open-label, phase 3, non-inferiority trial
Research in context Evidence before this study Patients with non-favourable risk acute myeloid leukaemia and a delayed response to intensive induction chemotherapy or relapsed acute myeloid leukaemia are candidates for allogeneic haematopoietic stem cell transplantation (HSCT).
Fludarabine/TBI 8 Gy versus fludarabine/treosulfan conditioning in patients with AML in first complete remission: a study from the Acute Leukemia Working Party of the EBMT | Bone Marrow Transplantation
Abstract The optimal reduced intensity conditioning (RIC) regimen is a matter of debate. We retrospectively compared conditioning with fludarabine plus fractionated total body irradiation of 8 Gy (FluTBI) and fludarabine plus treosulfan 30, 36 or 42 g/m2 (FluTreo) in 754 patients with AML above the age of 40 years undergoing an allogeneic hematopoietic stem cell transplant (HSCT) in first complete remission (CR).
Impact of conditioning regimen intensity on outcomes of second allogeneic hematopoietic cell transplantation for secondary acute myelogenous leukemia - Bone Marrow Transplantation
Abstract Limited data is available on factors impacting the outcomes of second hematopoietic cell transplantation (HCT2) in patients with secondary acute myeloid leukemia (sAML). This study aimed to assess HCT2 outcome for sAML comparing reduced-intensity (RIC) to myeloablative (MAC) conditioning. Two hundred and fifteen patients were included: RIC (n = 134), MAC (n = 81). The median follow-up was 41.1 (95% CI: 26.7–69.3) and 28.5 (95% CI: 23.9–75.4) months, respectively.
