Immunotherapies, such as checkpoint inhibitors, have transformed cancer treatment, however, their efficacy is largely restricted to tumors with an inflamed tumor microenvironment (TME). In contrast, immunologically cold tumors—such as pancreatic ductal adenocarcinoma (PDAC)—exhibit low levels of T cell infiltration, with T cells often excluded from the tumor parenchyma.
