mTORC1-TFEB/TFE3 signaling is associated with bile acid diversification during hepatic metabolic adaptation
Abstract INTRODUCTION RESULTS DISCUSSION MATERIALS AND METHODS Acknowledgments Supplementary Materials REFERENCES Information & Authors Metrics & Citations View Options References Figures Tables Media Share This article has 0 eLetter . Abstract The mechanistic target of rapamycin complex 1 (mTORC1) integrates nutrient and hormonal cues to regulate hepatic lipid metabolism with major implications for metabolic dysfunction-associated steatotic liver disease (MASLD).
Empagliflozin restores colonic fatty acid metabolism and ameliorates fibrotic signalling despite gut dysbiosis in Western diet-fed mice
Abstract Gut dysbiosis and an altered host colonic transcriptomic contribute to Western diet (WD) induced metabolic deterioration. In this pilot study we investigated the effect of empagliflozin on intestinal transcriptomics and microbiome. CL57BL/6J mice were fed a WD or control-diet with or without empagliflozin supplementation for 10 weeks. Colon and stool samples were used for transcriptomics and 16 S amplicon sequencing.
Association of the composition of the bone marrow tumor microenvironment in BCR::ABL1-negative myeloproliferative neoplasms with IFN-γ signaling and driver mutations | Leukemia
Abstract Constitutive JAK/STAT pathway activation is crucial in the pathogenesis of BCR::ABL1-negative myeloproliferative neoplasms (MPN), but has not yet been linked to interferon (IFN)-γ signaling and tumor microenvironment. Human JAK2 V617F-mutated cell lines, 265 bone marrow biopsies (BMB) of two MPN cohorts, and 50 non-neoplastic BMB, revealed an intrinsic activation of IFN-γ signaling, which was confirmed by public RNA expression data.
Neutralizing GDF-15 can overcome anti-PD-1 and anti-PD-L1 resistance in solid tumours - Nature

Abstract Cancer immunotherapies with antibodies blocking immune checkpoint molecules are clinically active across multiple cancer entities and have markedly improved cancer treatment1. Yet, response rates are still limited, and tumour progression commonly occurs2. Soluble and cell-bound factors in the tumour microenvironment negatively affect cancer immunity.

Inhibition of the Sterol Regulatory Element Binding Protein SREBF-1 Overcomes Docetaxel Resistance in Advanced Prostate Cancer
Resistance to antiandrogens and chemotherapy (Cx) limits therapeutic options for patients with hormone-sensitive and hormone-resistant prostate cancer (mCRPC).
Glucocorticoid treatment influences prostate cancer cell growth and the tumor microenvironment via altered glucocorticoid receptor signaling in prostate fibroblasts | Oncogene
Abstract Despite significant therapeutic advances in recent years, treatment of metastatic prostate cancer (PCa) remains palliative, owing to the inevitable occurrence of drug resistance. There is increasing evidence that epithelial glucocorticoid receptor (GR) signaling and changes in the tumor-microenvironment (TME) play important roles in this process.
Age-Dependent Surface Receptor Expression Patterns in Immature Versus Mature Platelets in Mouse Models of Regenerative Thrombocytopenia
1. Introduction Megakaryocytes are generated from hematopoietic stem cells in the bone marrow [1]. During maturation, they enlarge during endomitosis and accumulate RNA [2,3], ribosomes, and organelles [4,5]. Finally, proplatelet projections emerge, which release platelets into the circulation [6]. Newly formed platelets are rich in RNA and ribosomes, enabling them to synthesize proteins.

Proteins and antibodies HIS-tagged recombinant human Growth and Differentiation Factor-15 was produced in HEK293T cells (Invigate GmbH, Jena). The anti-human GDF-15 antibody used in this manuscript was generated in female C57BL/6J GDF-15-/–mice kindly provided by Dr. Jens Strelau (Heidelberg, Germany) and validated for binding affinity by surface plasmon resonance at NMI (Reutlingen, Germany). Specificity was tested by tissue cross-reactivity studies.

Downregulation of MTSS1 in acute myeloid leukemia is associated with a poor prognosis, chemotherapy resistance, and disease aggressiveness
Abstract Despite recent approval of targeted drugs for acute myeloid leukemia (AML) therapy, chemotherapy with cytosine arabinoside and anthracyclines remains an important pillar of treatment. Both primary and secondary resistance are frequent and associated with poor survival, yet the underlying molecular mechanisms are incompletely understood. In previous work, we identified genes deregulated between diagnosis and relapse of AML, corresponding to therapy naïve and resistant states, respectively.
Overcoming negative predictions of microRNA expressions to gemcitabine response with FOLFIRINOX in advanced pancreatic cancer patients
Pancreatic cancer (PC) is the third leading cause of cancer-related death in men and women in the USA and is anticipated to take the second place in 2020 ranking second after lung cancer [1]. In 2019, 45,750 PC deaths and 56,770 new cases were estimated in the USA alone; however, by 2030, 63,000 expected cancer death from PC are predicted, making it the number one cause of cancer-related death in the next 10 years [2].
Immunomodulation with pomalidomide at early lymphocyte recovery after induction chemotherapy in newly diagnosed AML and high-risk MDS
Abstract An immunosuppressive microenvironment promoting leukemia cell immune escape plays an important role in the pathogenesis of AML. Through its interaction with cereblon, a substrate receptor for the E3 ubiquitin ligase complex, pomalidomide leads to selective ubiquitination of transcription factors Aiolos and Ikaros thereby promoting immune modulation.
Next-generation computational tools for interrogating cancer immunity
Abstract The remarkable success of cancer therapies with immune checkpoint blockers is revolutionizing oncology and has sparked intensive basic and translational research into the mechanisms of cancer–immune cell interactions. In parallel, numerous novel cutting-edge technologies for comprehensive molecular and cellular characterization of cancer immunity have been developed, including single-cell sequencing, mass cytometry and multiplexed spatial cellular phenotyping.
SOCS2 is part of a highly prognostic 4-gene signature in AML and promotes disease aggressiveness - Scientific Reports
Abstract Acute myeloid leukemia (AML) is a heterogeneous disease with respect to its genetic and molecular basis and to patients´ outcome. Clinical, cytogenetic, and mutational data are used to classify patients into risk groups with different survival, however, within-group heterogeneity is still an issue.
