Protein Sample Preparation The full-length S. aureus scdA gene was inserted into the NcoI/EcoRI sites of the pET-28a expression vector (New England Biolabs). This vector adds an N-terminal six-His tag followed by a thrombin-cleavage site, yielding purified ScdA at ∼27 kDa with the tag ( Figure 1 A) and ∼25 kDa after tag removal ( Figure S1B ). A C-terminal domain-only (CTD, residues 64–224) variant was similarly cloned into pET-28a using the same procedure.
