To date, there has been no evidence for a basic tubulin residue playing a critical role in kinesin-based motility12, 13, 17. However, a recent study in yeast showed that introducing the CFEOM3-associated mutation of the TUBB3 basic residue (R262C or R262H) to the yeast TUB2 gene blocked Kip3p accumulation at MT plus-ends11. Therefore, we aimed to investigate whether this basic residue mutation could inhibit the in vitro motility of kinesin when expressed in TUBB3.
