Editor’s summary Acute chest syndrome (ACS) is one of the most severe complications that individuals with sickle cell disease (SCD) may experience. ACS, a form of acute lung injury, lacks any treatment option. Here, Chonat et al. found that the complement pathway is activated in individuals with SCD during ACS and in a mouse model of ACS. Deleting C3 or pharmacological inhibition of C5 alleviated disease severity in mice.
