FACS-based detection of extracellular ASC specks from NLRP3 inflammasomes in inflammatory diseases
The apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) is crucial for inflammasome assembly and activation of several inflammasomes, including NLRP3. ASC aggregates are detected in human sera post pyroptotic cell death, but their inflammasome origin remains unclear. This study aimed to develop a method to detect ASC aggregates originating from NLRP3 inflammasomes.

Inflammatory profile of lower risk myelodysplastic syndromes
INTRODUCTION There is increasing evidence that autoinflammation plays an important role in the pathogenesis of myelodysplastic syndromes (MDS), with several studies describing dysregulation of the innate immune response, as being particularly relevant.1 For example, Toll-like receptors (TLRs) have been shown to be over-expressed in haematopoietic stem/progenitor cells (HSPCs), compared to age-matched control subjects.2 Furthermore, in the HSPCs of MDS patients, there is enhanced activation of...
The effects of intrathecal rituximab on biomarkers in multiple sclerosis
Highlights • Despite intrathecal administration, rituximab was detectable in the serum but not the CSF. • CSF BAFF fell following rituximab administration, but serum BAFF increased. • Serum CD20+ and CD19+ cell populations fell in the peripheral blood compartment following rituximab administration. Abstract Objectives Clinical trials of IV-rituximab have proved successful. It is unclear whether intrathecal (IT)-rituximab is more efficacious at lower doses. We examine its effects on B-cell biomarkers.
