1 Introduction 9-Fluorenylmethyloxycarbonyl (Fmoc) solid-phase peptide synthesis (SPPS) is the gold standard method for the routine and rapid preparation of linear peptides <40 residues in length.[1-3] It involves the covalent attachment of the C-terminal amino acid of the peptide onto insoluble resin beads (typically consisting of polystyrene), followed by assembly via iterative coupling/deprotection cycles with Fmoc-amino acids.

Editor’s summary β cell antigen–specific CD4+ T cell responses promote autoimmune destruction of insulin-producing pancreatic β cells. The ability to easily quantify the extent of such responses could potentially benefit the management of patients with or at risk of type 1 diabetes. Lacorcia et al. show that autoreactive CD4+ T cell responses to C-peptide can be accurately measured by detecting secreted IL-2.
