Transplantation of endothelial progenitor cells attenuates pulmonary fibrosis via IFITM3-containing exosomes
Abstract Endothelial progenitor cells (EPCs) are critical for vascular regeneration after injury. However, their role in pulmonary fibrosis (PF) remains unclear. Here, we identified a previously unrecognized population of lung capillary EPCs coexpressing c-KIT and FOXF1 (capEPCs) in adult human and mouse lungs. capEPCs were significantly reduced in lungs from patients with PF and in bleomycin-injured mice, implicating loss of this regenerative endothelial population in disease pathogenesis.
Developing Highly Effective Nanoparticle mRNA Therapeutic for Pediatric Acute Respiratory Distress Syndrome
1 Introduction Pediatric acute respiratory distress syndrome (PARDS), a life-threatening complication of acute lung injury (ALI) in children, remains a major cause of morbidity and mortality [1,2]. Mortality rates are significantly higher in children who develop ARDS following critical injury (34.0%) than those who do not (1.7%) [3]. In severe cases, mortality can reach 50% [4].
Precision nanomedicine for pulmonary diseases: from molecular targeting to clinical translation
Abstract The pulmonary system is a vital interface between the body and the external environment, making it highly vulnerable to environmental, infectious, and genetic insults. Precision nanomedicine offers a promising strategy to overcome the limitations of conventional gene and drug therapies, including safety concerns associated with viral vectors, instability of therapeutic agents, suboptimal cellular internalization, and a critical lack of tissue- and cell-specific targeting.
Emergence of fibrotic pericytes and their transcriptional regulation in pulmonary fibrosis
Abstract Multiple cell types have been implicated in pathogenesis of pulmonary fibrosis with pericytes emerging as a new focus due to their role in promoting fibrotic remodeling. Fibrotic environment changes normal pericyte functions, but transcriptional programs regulating pericyte transition towards fibrotic state remain unclear.
FOXF2 regulates pericyte–endothelial signaling required for vascular homeostasis after neonatal hyperoxic lung injury - Nature Communications
Abstract Pulmonary vascular development is essential for alveolarization, and disruption of this process contributes to pathogenesis of bronchopulmonary dysplasia (BPD). Proper vascular development requires an orchestration of many cell types within the lung. However, the transcriptional mechanisms by which pericytes support the endothelium in the postnatal lung remain poorly understood.
Engineering a SARS-CoV-2 vaccine targeting the RBD cryptic-face via immunofocusing
Abstract The receptor-binding domain (RBD) of the SARS-CoV-2 spike protein is the main target of neutralizing antibodies. Although they are infrequently elicited during infection or vaccination, antibodies that bind to the conformation-specific cryptic face of the RBD display remarkable breadth of binding and neutralization across Sarbecoviruses.
