Simple generation of cleavable labels for multiplexed imaging We present a simple and inexpensive approach for the amine-targeted labeling of biomolecules with fluorophores so that the linker between them can be easily cleaved by a mild reducing agent. By providing a convenient way to introduce cleavable linkers, our method can simplify high-content fluorescence imaging based on repeated staining and imaging cycles. 1 Introduction During embryonic development and organogenesis, differentiating cells exit the cell cycle and enter a prolonged or permanent G0 growth arrest to achieve their final mature state, which represents the most common cell state in adult organs.[1, 2] The transition from proliferation to cell cycle exit is accompanied by a transcriptional and functional switch in essential regulators of the cell cycle program.
