Review: Earthbound by Katee Stein — SFF Insiders
At the heart of Earthbound are the central trio of characters, Knox, Tahren, and Emilia, who all undergo deeply embedded traumas, and find solace in both memory, and each other throughout the story. Responsive Antibody Conjugates Enable Quantitative Determination of Intracellular Bond Degradation Rate
Highlights • OligoTEA-based FRET probes enabled visualization of intracellular bond cleavage • Mass-action kinetics can reliably model and quantitate intracellular processes • The disulfide bond half-life in the HER2 pathway was determined (t 1/2 = 92 min) • This study highlights the pitfalls of measuring intracellular rates in cell-free assays Summary Degradable crosslinkers that respond to intracellular biological stimuli are a critical component of many drug delivery systems.
Continuous Solids Discharging centrifugation to Solve Clarifying High-Cell-Density Mammalian Cell Cultures
Clarification is typically the first unit operation in the purification of monoclonal antibodies (MAbs) and other proteins from mammalian cell cultures. This process removes cells and cellular debris from the culture fluid to produce a clarified cell culture supernatant that will be suitable for further purification (1). Anti‐α4 Integrin Antibody Blocks Monocyte/Macrophage Traffic to the Heart and Decreases Cardiac Pathology in a SIV Infection Model of AIDS
Methods and Results We directly blocked monocyte/macrophage traffic to the heart in an SIV model of AIDS using an anti‐alpha‐4 integrin antibody (natalizumab). Nineteen Rhesus macaques were SIVmac251 infected and CD8‐lymphocyte depleted for the development of rapid AIDS. Ten animals received natalizumab once a week, for 3 weeks, and were sacrificed 1 week later.
