Primary Schwann cell culture Primary Schwann cell cultures were prepared from sciatic nerves dissected from PND 10–15 mice (n = 4 per genotype) under aseptic conditions using a previously published protocol (Weinstock et al., 2020). Briefly, the mice were sacrificed by the inhalation of carbon dioxide and the removed sciatic nerves were placed in Leibovitz L-15 medium, unsheathed of epineurium, and then transferred to high-glucose DMEM supplemented with 0.25% dispase II and 0.05% collagenase.
