Myeloproliferative neoplasms (MPN) are a subset of clonal stem cell disorders, including essential thrombocythemia (ET), polycythemia vera (PV), and primary myelofibrosis. Somatic mutations in JAK2, CALR, and MPL in hematopoietic stem cells are primary drivers of MPN pathogenesis. Conversely, JAK2 germline variants have been reported in two distinct contexts. Certain rare germline variants, such as those described in familial MPN, are sufficient on their own to cause disease [1].
