Automated and streamlined manufacturing of TIL therapies
Live30 webinars are thirty-minute presentations designed to update you on the latest innovations, applications, and data in a fast yet interactive format. The use of tumor-infiltrating T cells (TILs) for cancer therapy has shown some tremendous results, leading to the first FDA approval in early 2024. Yet, the journey towards clinical realization presents some important challenges that demand innovative solutions.
The effect of a single dose of nivolumab prior to isolated limb perfusion for patients with in-transit melanoma metastases: An interim analysis of a phase Ib/II randomized double-blind placebo-controlled trial (NivoILP trial)
ILP has shown to achieve high response rates in patients with melanoma ITM. Possibly there is a synergistic mechanism of action of ILP and anti-PD1. The aim of this trial was to investigate the safety and efficacy of adding a single dose of systemic anti-PD1 to isolated limb perfusion (ILP) for patients with melanoma in-transit metastases (ITM).
Targeted Therapy in the Palliative Setting of Colorectal Cancer-Survival and Medical Costs
Abstract : Simple Summary Targeted therapy targeting the EGFr and vascular endothelial growth factor (VEGF) is often combined with chemotherapy in palliative treatment of metastatic colorectal cancer. The aim of this population-based cohort study was to examine factors influencing treatment decisions for targeted therapy and their effect on overall survival in palliative metastatic colorectal cancer.
Prognostic significance of sentinel lymph node biopsy in thick primary melanomas (>4mm)
Background: The key prognostic factors for staging patients with primary cutaneous melanoma are Breslow thickness, ulceration, and sentinel lymph node biopsy (SLNB) status. The MSLT-I trial verified SN status as the most important prognostic factor in patients with intermediate thickness melanoma (Breslow thickness 1-4mm).

Phase II LEAP-004 Study of Lenvatinib Plus Pembrolizumab for Melanoma With Confirmed Progression on a Programmed Cell Death Protein-1 or Programmed Death Ligand 1 Inhibitor Given as Monotherapy or in Combination | Journal of Clinical Oncology
Abstract Effective treatments are needed for melanoma that progresses on inhibitors of programmed cell death protein-1 (PD-1) or its ligand (PD-L1). We conducted the phase II LEAP-004 study to evaluate the combination of the multikinase inhibitor lenvatinib and the PD-1 inhibitor pembrolizumab in this population (ClinicalTrials.gov identifier: NCT03776136).
3. Results A total of 287 patients with melanoma ITM from 21 institutions in eight different countries treated with immunotherapy were included in the analysis ( Supplementary Table 1 ). Overall, 161 patients (56%) were male, and the median age at the start of ICI was 69 years (IQR 59–78). The median time from melanoma diagnosis to the first in-transit recurrence was 13 months (IQR 5–30).
Surgery of melanoma metastases after systemic therapy - the SUMMIST trial
Carl Jacob Holmberg Dimitrios Katsarelias Henrik Jespersen Lars Ny Roger Olofsson Bagge Background: PD1-inihibitors are currently used as first-line treatment for metastasized malignant melanoma. The response rate is over 40% with a complete response (CR) rate of up towards 20%. In patients with partial response (PR) or stable disease (SD), there is no clear standard for second line treatment, however a common strategy is to continue with PD-1 inhibition up to two years.
Mouse avatars take off as cancer models
We think your discussion on the use of mice with human tumours as cancer models is too pessimistic (Nature 560, 156–157; 2018). These mouse ‘avatars’ can now be armed with human immune cells and are already providing promising insights into immunotherapies (Y. Choi et al. Exp. Mol. Med. 50, 99; 2018). One example is a personalized mouse model we developed for melanoma.
