A case of Bundibugyo virus disease treated with monoclonal antibodies and remdesivir
Abstract Clinical data on Bundibugyo virus (BDBV) infection are limited, and no approved virus-specific treatment exists. We describe the case of a previously healthy 39-year-old healthcare worker who acquired BDBV infection while working in Ituri Province, Democratic Republic of the Congo (DRC), and was medically evacuated to Germany.
Sustained HIV-1 remission after heterozygous CCR5Δ32 stem cell transplantation - Nature
Abstract HIV cure is exceptionally rare, documented in only six cases among the estimated 88 million individuals who have acquired HIV since the epidemic's onset1–6. Successful cures, including the pioneering Berlin patient, are limited to individuals receiving allogeneic stem cell transplants (allo-SCT) for hematological cancers.
The cellular and molecular cardiac tissue responses in human inflammatory cardiomyopathies after SARS-CoV-2 infection and COVID-19 vaccination - Nature Cardiovascular Research
Abstract Myocarditis, characterized by inflammatory cell infiltration, can have multiple etiologies, including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection or, rarely, mRNA-based coronavirus disease 2019 (COVID-19) vaccination. The underlying cellular and molecular mechanisms remain poorly understood.
Gut microbiota dysbiosis is associated with altered tryptophan metabolism and dysregulated inflammatory response in COVID-19 - npj Biofilms and Microbiomes
Abstract The clinical course of COVID-19 is variable and often unpredictable. To test the hypothesis that disease progression and inflammatory responses associate with alterations in the microbiome and metabolome, we analyzed metagenome, metabolome, cytokine, and transcriptome profiles of repeated samples from hospitalized COVID-19 patients and uninfected controls, and leveraged clinical information and post-hoc confounder analysis.
Associations of myeloid cells with cellular and humoral responses following vaccinations in patients with neuroimmunological diseases - Nature Communications
Abstract Disease-modifying therapies (DMTs) are widely used in neuroimmunological diseases such as multiple sclerosis (MS) and neuromyelitis optica spectrum disorder (NMOSD). Although these treatments are known to predispose patients to infections and affect their responses to vaccination, little is known about the impact of DMTs on the myeloid cell compartment.
CD4+ T cell calibration of antigen-presenting cells optimizes antiviral CD8+ T cell immunity - Nature Immunology
|
Abstract Antiviral CD8+ T cell immunity depends on the integration of various contextual cues, but how antigen-presenting cells (APCs) consolidate these signals for decoding by T cells remains unclear. Here, we describe gradual interferon-α/interferon-β (IFNα/β)-induced transcriptional adaptations that endow APCs with the capacity to rapidly activate the transcriptional regulators p65, IRF1 and FOS after CD4+ T cell-mediated CD40 stimulation.

Distinct tissue niches direct lung immunopathology via CCL18 and CCL21 in severe COVID-19 - Nature Communications
|
Abstract Prolonged lung pathology has been associated with COVID-19, yet the cellular and molecular mechanisms behind this chronic inflammatory disease are poorly understood. In this study, we combine advanced imaging and spatial transcriptomics to shed light on the local immune response in severe COVID-19. We show that activated adventitial niches are crucial microenvironments contributing to the orchestration of prolonged lung immunopathology.

Sensing Microbial Viability through Bacterial RNA Augments T Follicular Helper Cell and Antibody Responses
Highlights • Innate detection of bacterial RNA directs Tfh differentiation and antibody responses • TRIF-dependent IRF3 and inflammasome pathways orchestrate the Tfh response • CX3CR1+CCR2– monocytes secrete IL-1β in response to live bacteria and drive Tfh cells • IFN-β-dependent secretion of IL-1β engages T cell IL-1R1 to mediate Tfh differentiation Summary Live vaccines historically afford superior protection, yet the cellular and molecular mechanisms mediating protective immunity remain...
