| Cell Chemical Biology Verified
Highlights • Top1-targeting camptothecin drugs induce oxidative stress and lipid peroxidation • Lipid-derived electrophiles modify a Top1 active-site cysteine by Michael addition • Electrophile-modified Top1 forms covalent complexes with cleaved DNA • Oxidative stress may mediate cytotoxicity of Top1 poisons in vivo Summary Topoisomerase 1 (Top1) reversibly nicks chromosomal DNA to relax strain accumulated during transcription, replication, chromatin assembly, and chromosome condensation.
