What is already known? NOS1 (neuronal) worsens ischaemic brain injury, whereas NOS3 (endothelial) is protective in experimental stroke. NO inhibits mitochondrial complex IV and reduces oxygen consumption. What does this study add? During OGD/R, NOS inhibition in both endothelial cells and neurons inhibits NOS uncoupling-derived ROS generation. NOS inhibition protects endothelial cells and neurons against OGD/R injury by paradoxically reducing mitochondrial respiration.
