Intestinal inflammation promotes neuroinflammation and PD-associated nigrostriatal pathology independently of LRRK2 G2019S kinase activity

Abstract Introduction: The pathogenesis of Parkinson’s disease (PD) has been linked to environmental factors, toxins, genetics, and peripheral inflammation. Importantly, intestinal inflammation like that seen in Crohn’s disease (CD) or food allergies has been implicated in risk for neurodegeneration and late-onset PD. Further, CD and PD share genetic risk factors including gain-of-function leucine-rich repeat kinase 2 (LRRK2) mutations.

Challenges in harmonizing inflammatory markers in AD/ADRD Consensus Statement: Standard reporting of CSF inflammatory marker assay validation and performance will improve incorporation of inflammatory markers into AT(N) scheme. Recommendation: A minimum set of CSF inflammatory markers should be measured by established laboratories for accuracy and precision using assays from manufacturers committed to transparency and continuous improvement.

Samples from the National Centralized Repository for Alzheimer’s Disease and Related Dementias (NCRAD) were used. Cryopreserved peripheral blood mononuclear cells (PBMCs) from GRN-FTD and age- and sex-matched neurologically healthy controls (n = 25 per participant group) were received and kept in liquid nitrogen cryostorage until use. See Tables 1 and 2, and Supplementary File 1 for participant demographics.
