Engineered organoid platforms for mechanistic studies of gut microbiota-cancer interactions
Abstract Associations between the gut microbiota and cancer are increasingly reproducible at the cohort level, but association alone cannot establish whether a microorganism, microbial product, or metabolite directly alters tumor-relevant host biology. Engineered organoid systems address this gap by placing defined microbial exposures against human or genetically specified epithelia under experimentally controlled conditions.
Validating FR20 and D-FR20 for Bromodomain and Extra-Terminal Domain Inhibitor Resistance in Triple-Negative Breast Cancer
To the Editor, We read with interest the study by Liu et al. [1], which combines time-series single-cell RNA sequencing, perturbational transcriptomics, and functional experiments to examine resistance to bromodomain and extra-terminal domain inhibitors in triple-negative breast cancer.

Clarifying causal and translational inference in studies of ambient PM2.5 and lung cancer
To the Editor, We read with interest the nationally representative case–control study by Alhattab et al. reporting an association between long-term ambient PM2.5 exposure and lung cancer risk in a relatively low-pollution setting using high-resolution geospatial modelling [1]. As lung cancer among never-smokers becomes increasingly prominent and environmental exposures assume a larger role in prevention frameworks, these findings are timely.

Integrated fecal SDC2 methylation and serum tumor markers improve noninvasive detection of colorectal cancer
Abstract Background: Fecal SDC2 methylation is a promising noninvasive biomarker for colorectal cancer (CRC), whereas serum tumor markers, including carcinoembryonic antigen (CEA), carbohydrate antigen 19-9 (CA19-9), CA242, and CA50, are primarily used for postoperative surveillance and have limited value as standalone early diagnostic markers. This study evaluated the diagnostic performance of combining fecal SDC2 methylation with these four serum markers for CRC detection.
Skeletal-muscle-targeted non-viral delivery of full-length DMD mRNA for Duchenne muscular dystrophy
|
Abstract Duchenne muscular dystrophy (DMD) is a severe, progressive muscle-wasting disorder caused by mutations in the DMD gene, which encodes dystrophin. Although gene therapy using viral vectors has shown promise for the treatment of DMD, the clinical application of viral gene therapies is limited by vector toxicity, immunogenicity and the inability to package full-length dystrophin.

GLP-1 RA Therapy After Bariatric Surgery: Phenotype, Strategy and Success
To the Editor, We read with great interest the target trial emulation by Wu et al. [1], which provides timely Chinese real-world evidence that GLP-1 receptor agonist initiation may promote clinically meaningful weight loss among suboptimal responders after metabolic bariatric surgery. This work is particularly relevant as incretin-based therapies are increasingly extending beyond primary obesity management into post-bariatric rescue and maintenance care.

Reframing molecular superiority in the differential diagnosis of eczema and psoriasis
Dear Editor, We read with interest the study by Schmitt et al., which compared dermatopathology with NOS2/CCL27-based PCR for differentiating eczema from psoriasis.1 As biologic therapies increasingly depend on precise inflammatory endotyping, improving diagnostic discrimination between these two entities has become a clinically important priority. In this context, the reported molecular advantage is noteworthy, but several interpretive issues warrant clarification before routine implementation.

The relationship between magnetic resonance imaging (MRI) feature and symptom remains unclear. Objective To evaluate the performance of MRI radiomics model for differentiating symptom heterogeneity of extrinsic adenomyosis, pain, abnormal uterine bleeding (AUB), infertility, and no symptom.
In Situ Programming of the Tumor Microenvironment to Alleviate Immunosuppression for Pancreatic Cancer Immunotherapy
1 Introduction Over the past few years, STING (stimulator of interferon genes) receptors have emerged as pivotal activators of both innate and adaptive immune responses against cancer through the activation of the interferon (IFN) signaling pathway.[1] A common therapeutic strategy for leveraging the STING pathway involves the direct delivery of agonists to tumor sites.
