The SPPS process occurs in a series of semibatch reactors, following Fmoc chemistry, with a DIC coupling agent and the Oxyma additive in a DMF solvent and with piperidine used for the deprotection step. While there are promising new methodologies for the SPPS process that include extra reactors, we focus here on the common setup with two reactors that are repeatedly utilized during each cycle of the build.
