Abstract Opioids are among the most widely prescribed treatments for pain; however, prolonged use leads to adverse effects, including reduced analgesic efficacy (tolerance) and paradoxically heightened pain sensitivity (opioid-induced hyperalgesia, OIH). Neurons that detect noxious stimuli within the dorsal root ganglion (DRG), referred to as nociceptors, mediate both the beneficial and maladaptive effects of opioids.

Our machine learning algorithms show few species differences between mouse and human DRG neuron electrophysiology under baseline conditions. These are important findings for the study of neuronal excitability in the context of pain therapeutic development.
