Significance Nicotine addiction affects one-third of the population and is the leading cause of preventable death. The habenula, an evolutionarily conserved brain structure, is enriched in nicotinic acetylcholine receptors (nAChRs) and critically controls nicotine intake. We found that GPR151, an orphan GPCR with selective expression in rodent and human habenular neurons, specifically regulates cyclic adenosine monophosphate levels and synaptic neurotransmission.
