Mitochondrial dynamics in orthoflavivirus infection: insights from human and mosquito hosts
Abstract Mosquito-borne orthoflaviviruses alternate between vertebrates and mosquitoes, where they establish distinct infection outcomes. In humans, infection is acute and associated with immune activation and cellular damage, whereas in mosquitoes, viruses persist lifelong without overt pathology. Mitochondria, as central regulators of metabolism, signaling, and immunity are emerging as key determinants of these distinct responses.
The Hsf1-sHsp cascade has pan-antiviral activity in mosquito cells - Communications Biology
Abstract Aedes mosquitoes transmit pathogenic arthropod-borne (arbo) viruses, putting nearly half the world’s population at risk. Blocking virus replication in mosquitoes is a promising approach to prevent arbovirus transmission, the development of which requires in-depth knowledge of virus-host interactions and mosquito immunity.
Characterization of gene regulatory elements and dynamic antimicrobial immune responses in mosquito cells using PRO-seq
Abstract The Aedes aegypti mosquito transmits arboviruses such as dengue, Zika, and chikungunya virus, posing a substantial threat to global health. The mosquito immune response determines virus transmission, yet, insight into the transcriptional regulation of mosquito immunity remains limited. In this study, we optimized the nascent RNA-sequencing method Precision Run-On sequencing (PRO-seq) for Aedes aegypti Aag2 cells.
A satellite repeat-derived piRNA controls embryonic development of Aedes
Abstract Tandem repeat elements such as the diverse class of satellite repeats occupy large parts of eukaryotic chromosomes, mostly at centromeric, pericentromeric, telomeric and subtelomeric regions1. However, some elements are located in euchromatic regions throughout the genome and have been hypothesized to regulate gene expression in cis by modulating local chromatin structure, or in trans via transcripts derived from the repeats2,3,4.
Crossing the Mucosal Barrier: A Commensal Bacterium Gives Dengue Virus a Leg-Up in the Mosquito Midgut
Commensal bacteria that colonize the midgut of Aedes aegypti mosquitoes can influence the transmission of arthropod-borne viruses. In this issue of Cell Host & Microbe, Wu et al. (2019) Wu P. Sun P. Nie K. Zhu Y. Shi M. Xiao X. Liu H. Liu Q. Zhao T. Chen X. et al. A gut commensal bacterium promotes mosquito permissiveness to arboviruses. Cell Host Microbe.
