Atlas of cell types and regulatory elements underlying human facial diversity
Abstract Human genetic diversity generates an astonishing variety of facial shapes, and craniofacial anomalies rank among the most common birth defects. Identifying the cellular mechanisms that mediate the genome’s influence on facial variation remains a challenge. Here we created a multimodal facial atlas across embryonic weeks 6–11, providing single-cell transcriptomics, chromatin accessibility and spatial transcriptomics, all at single-cell resolution.
Three-dimensional facial gestalt analysis for three neurodevelopmental disorders: Koolen-de Vries, Jansen-de Vries and KBG syndrome
Abstract Diagnosing children with developmental disorders is often challenging due to the large number of rare syndromes and their variable clinical presentations. While next-generation sequencing has improved diagnostic yield, results are frequently inconclusive, highlighting the continued importance of detailed phenotypic assessment.

Comment on “De Novo Reconstruction of 3D Human Facial Images from DNA Sequence”
1 Main Text The emergence of AI-driven DNA-based facial reconstruction technologies marks a significant milestone in the intersection of genomics, computer vision, and forensic science. Among these, the Difface model introduced by Jiao et al.[1] represents a notable advancement, leveraging contrastive learning and diffusion models to generate three-dimensional (3D) facial images from genetic data.
Novel multi-omics deconfounding variational autoencoders can obtain meaningful disease subtyping
Abstract In the field of precision medicine, the use of multi-omics data for patient stratification holds great promise for delivering tailored treatments based on comprehensive individual biological profiles. The clinical potential of (multi-)omics data, however, faces significant limitations due to the presence of confounding factors in the data, such as noise from experimental procedures or other irrelevant biological signals.

Initial Steps towards a Multilevel Functional Principal Components Analysis Model of Dynamical Shape Changes
Abstract : In this article, multilevel principal components analysis (mPCA) is used to treat dynamical changes in shape. Results of standard (single-level) PCA are also presented here as a comparison. Monte Carlo (MC) simulation is used to create univariate data (i.e., a single “outcome” variable) that contain two distinct classes of trajectory with time.
Precise modulation of transcription factor levels identifies features underlying dosage sensitivity - Nature Genetics
|
Abstract Transcriptional regulation exhibits extensive robustness, but human genetics indicates sensitivity to transcription factor (TF) dosage. Reconciling such observations requires quantitative studies of TF dosage effects at trait-relevant ranges, largely lacking so far.

Genetic variants underlying differences in facial morphology in East Asian and European populations - Nature Genetics
|
Abstract Facial morphology—a conspicuous feature of human appearance—is highly heritable. Previous studies on the genetic basis of facial morphology were performed mainly in European-ancestry cohorts (EUR). Applying a data-driven phenotyping and multivariate genome-wide scanning protocol to a large collection of three-dimensional facial images of individuals with East Asian ancestry (EAS), we identified 244 variants in 166 loci (62 new) associated with typical-range facial variation.

Facial Genetics: A Brief Overview
Introduction The facial surface is readily visible and identifiable with a close relationship to the underlying cartilaginous and skeletal structures (Stephan et al., 2005; Wilkinson et al., 2006; De Greef et al., 2006; Al Ali et al., 2014b; Shrimpton et al., 2014).
Rapid neural categorization of angry and fearful faces is specifically impaired in boys with autism spectrum disorder
Abstract Background Difficulties with facial expression processing may be associated with the characteristic social impairments in individuals with autism spectrum disorder (ASD). Emotional face processing in ASD has been investigated in an abundance of behavioral and EEG studies, yielding, however, mixed and inconsistent results.

Modeling 3D Facial Shape from DNA
Human facial diversity is substantial, complex, and largely scientifically unexplained. We used spatially dense quasi-landmarks to measure face shape in population samples with mixed West African and European ancestry from three locations (United States, Brazil, and Cape Verde). Using bootstrapped response-based imputation modeling (BRIM), we uncover the relationships between facial variation and the effects of sex, genomic ancestry, and a subset of craniofacial candidate genes.
