Pre–CAR-T B-cell–directed immunotherapy exposure and outcomes after CD19 CAR-T for large B-cell lymphoma: An EBMT registry analysis
CD19-directed chimeric antigen receptor T-cell therapy (CAR-T) is standard-of-care second-line treatment for relapsed or refractory large B-cell lymphoma (LBCL) [1,2,3]. Bispecific antibodies (BSA) and polatuzumab vedotin (pola) are increasingly used prior to CAR-T, but the impact of such exposure, particularly before leukapheresis, when therapies may affect T-cell collection and fitness, remains unclear [4,5,6,7].

While about 60–70% of patients with DLCBL can be cured with R-CHOP chemoimmunotherapy, 30–40% relapse or are refractory to treatment.

Chimeric antigen receptor T-cell therapy is equally effective and safe in older (≥70) and younger patients with multiple myeloma: a multicenter retrospective analysis
|
Dear Editor, The introduction of chimeric antigen receptor T-cell (CAR-T) therapies has significantly transformed the treatment landscape for patients with relapsed/refractory (r/r) multiple myeloma (MM). BCMA-directed CAR-T cell therapies, idecabtagene vicleucel (ide-cel) and ciltacabtagene autoleucel (cilta-cel), have revolutionized the field by demonstrating remarkable efficacy in pivotal clinical trials [1, 2].

Immune reconstitution in AML and MDS patients undergoing allogeneic hematopoietic cell transplantation (allo-HCT) with treosulfan- or TBI-based conditioning | Bone Marrow Transplantation
To the Editor: Cellular and humoral immune reconstitution after allo-HCT impacts infections, graft-versus-host disease (GvHD) and relapse-risk. How conditioning regimens affect immune recovery is largely unknown. A study comparing fludarabine/treosulfan (FluTreo) and fludarabine/total body irradiation (FluTBI) conditioning regimens showed no significant differences in overall survival (OS) and relapse-free survival (RFS) or GvHD incidence [1].
Hematopoietic stem cell transplantation for DLBCL: a report from the European Society for Blood and Marrow Transplantation on more than 40,000 patients over 32 years | Blood Cancer Journal
Abstract Autologous(auto-) and allogeneic(allo-) hematopoietic stem cell transplantation (HSCT) are key treatments for relapsed/refractory diffuse large B-cell lymphoma (DLBCL), although their roles are challenged by CAR-T-cells and other immunotherapies. We examined the transplantation trends and outcomes for DLBCL patients undergoing auto-/allo-HSCT between 1990 and 2021 reported to EBMT.
Comparable outcomes for TBI-based versus treosulfan based conditioning prior to allogeneic hematopoietic stem cell transplantation in AML and MDS patients | Bone Marrow Transplantation
Abstract Allogeneic hematopoietic stem cell transplantation (allo-HCT) is a standard treatment for patients with AML and MDS. The combination of fractionated total body irradiation(8GyTBI/Flu) with fludarabine is an established conditioning regimen, but fludarabine/treosulfan(Flu/Treo) constitutes an alternative in older/comorbid patients.
Allogeneic hematopoietic stem cell transplantation for NK/T-cell lymphoma: an international collaborative analysis - Leukemia
Abstract Natural killer/T-cell lymphomas (NKTCL) represent rare and aggressive lymphoid malignancies. Patients (pts) with relapsed/refractory disease after Asparaginase (ASPA)-based chemotherapy have a dismal prognosis. To better define the role of allogeneic hematopoietic stem cell transplantation (allo-HSCT), we conducted a retrospective analysis of data shared with the European Society for Blood and Marrow Transplantation (EBMT) and cooperating Asian centers.
