Fine-tuning of polygenic score to local data improves risk prediction and clinical implementation
Abstract Polygenic risk scores (PRS) hold promise for stratifying inherited disease risk, yet their clinical adoption remains constrained by poor transferability across populations and cohorts. To bridge this gap, we propose a pragmatic framework that recalibrates externally derived PRSs to local datasets through machine learning-based optimization.
Genetic evidence supports the combined targeting of lipoprotein(a) and LDL cholesterol to reduce coronary artery disease risk
|
Abstract Distinct genetic mechanisms govern how lipoprotein(a) (Lp(a)) and low-density lipoprotein cholesterol (LDL-C) promote atherosclerosis. It remains unclear whether targeting both provides additive cardiovascular benefits. Here we use coding loss-of-function variants in LPA and PCSK9 and genetic scores associated with Lp(a) and LDL-C levels to evaluate the effects of lowering Lp(a) and LDL-C on coronary artery disease (CAD) risk.

Prevalence and effects on outcomes of intracranial tumors and structural alterations in children with central precocious puberty and early and fast puberty
Abstract Objective: To investigate intracranial lesion prevalence in children with central precocious puberty (CPP) and early and fast puberty (EFP), and assess impacts on progression and GnRHa response. Methods: This retrospective study included 1,087 CPP/EFP patients (2011–2023) who underwent brain MRI. Patients were stratified by MRI findings: no lesion, other (incidental) lesions, and pathogenic lesions (hypothalamic hamartoma/glioma).

Application of slow-controlled release fertilizer coordinates the carbon flow in carbon-nitrogen metabolism to effect rice quality
Abstract Slow-controlled release fertilizers are experiencing a popularity in rice cultivation due to their effectiveness in yield and quality with low environmental costs. However, the underlying mechanism by which these fertilizers regulate grain quality remains inadequately understood.
