GVHD prophylaxis with PTCy, ATG, PTCy+ATG, or no in vivo T-cell depletion in RIC matched-donor PBSC transplantation for AML and MDS
Abstract Post-transplant cyclophosphamide (PTCy) has transformed graft-versus-host disease (GVHD) prophylaxis but its role in matched-donor reduced-intensity allogeneic hematopoietic cell transplantation (allo-HCT) remains uncertain.
ALLOPLUS: allogeneic hematopoietic stem cell transplantation in children and adult people living with human immunodeficiency virus, a retrospective study of the SFGM-TC
|
The incidence rate of malignancies is increased in people living with HIV (PLWH) [1]. The first allogeneic hematopoietic stem cell transplantation (alloHSCT) in a human immunodeficiency virus (HIV)-seropositive recipient suffering from a hematological disorder was performed in 1989 [2], two years after zidovudine received approval for HIV treatment.

Fludarabine, cyclophosphamide and 2 Gy TBI with PT-Cy as a single platform for haplo and unrelated donor allo-SCT in older AML or MDS patients
|
Abstract Allogeneic stem-cell transplantation (allo-SCT) is met with increased toxicity and relapse in elderly patients with acute myeloid leukemia (AML) or myelodysplastic syndrome (MDS). The nonmyeloablative conditioning (NMAC) regimen combining 2GyTBI, fludarabine, and cyclophosphamide (FluCyTBI) with post-transplant cyclophosphamide (PTCy) has been described as well adapted to AML/MDS patients aged ≥70yo in the haplo-identical setting.

Allogeneic hematopoietic stem cell transplantation in elderly patients with AML and MDS: navigating risks and optimizing outcomes
Abstract The growing burden of acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS) among aging populations underscores the critical need for tailored treatment strategies for elderly patients. While allogeneic hematopoietic stem cell transplantation (allo-HSCT) remains the only potentially curative option for eligible patients, its application in older adults is fraught with significant challenges.
Impact of stem cell source in HLA-matched hematopoietic stem cell transplant for acute lymphoblastic leukemia in first complete remission: a study from the GRAALL and the SFGM-TC
|
In myeloablative HLA-matched hematopoietic stem cell transplantation (HSCT) peripheral blood stem cells (PBSC) increase the risk of chronic graft-versus-host disease (cGVHD) compared to bone marrow (BM) [1, 2]. While cGVHD is associated with substantial morbidity, it may also augment graft-versus-leukemia activity.

Alternative versus matched unrelated donor in adolescents and young adults transplanted for acute myeloid leukemia in first complete remission: A report from the SFGM-TC
|
Allogeneic hematopoietic stem cell transplantation (HSCT) is considered the standard of care for adults with high-risk acute myeloid leukemia (AML) in first complete remission (CR1). In patients with intermediate-risk AML, the decision to proceed with HSCT in CR1 should be carefully weighed against the predicted risk of relapse and the anticipated treatment-related mortality.

Use of bone marrow cells is associated with improved outcomes when compared to peripheral blood stem cell after haplo-identical transplants with post transplant cyclophosphamide, a study from of the CTIWP-EBMT | Bone Marrow Transplantation
Abstract Few studies analyzed the impact of bone marrow (BM) or peripheral blood stem cells (PBSC) on outcomes after haploidentical transplantation using post-cyclophosphamide (Haplo-PTCY). We analyzed 8854 adults with malignant disorders, given a first Haplo-PTCY. BM cells was used in 2914 and PBSC in 5940 patients. Multivariate models were built adjusting for the statistical differences between the 2 groups.
Thiotepa-busulfan-fludarabine compared to clofarabine-based conditioning for haploidentical transplant with posttransplant cyclophosphamide in patients with myeloid malignancies: a retrospective study from the SFGM-TC | Bone Marrow Transplantation
Abstract The optimal reduced-intensity conditioning (RIC) regimen for haploidentical hematopoietic stem cells transplantation (haplo-HSCT) using post-transplant cyclophosphamide as graft-versus-host disease (GVHD) prophylaxis has yet to be determined. Potential RIC regimen for haplo-HSCT in myeloid malignancies include Clofarabine-Baltimore (CloB) and TBF (thiotepa-busulfan-fludarabine). This multicenter retrospective study compared 297 adult patients receiving CloB (n = 59) or TBF (n = 238).
Venetoclax and azacitidine for molecular relapse after intensive chemotherapy in NPM1 or CBF AML: a FILO study - Blood Cancer Journal
Dear Editor, In acute myeloid leukemia (AML), molecular response assessment and sequential follow-up by real-time quantitative PCR (RT-qPCR) during remission are routinely used in patients with NPM1 mutations (NPM1mut), RUNX1::RUNX1T1, or CBFB::MYH11 transcripts [1, 2].

Venetoclax–Azacitidine Versus Azacitidine for the Treatment of Primary Refractory or First Relapsed Acute Myeloid Leukemia. An IPC‐DATAML‐MSKCC Retrospective Study
To the Editors, Relapsed or refractory (R/R) AML have a poor outcome when treated with intensive chemotherapy (≈30% response rate) [1], or azacitidine (AZA, ≈20% response rate) [2-5]. Both strategies are associated with a poor overall survival (OS) ranging between 6 and 9 months. Venetoclax (VEN)–AZA has been approved for newly-diagnosed AML patients ineligible for IC [6].

Transplantation for myelofibrosis patients in the ruxolitinib era: a registry study from the Société Francophone de Greffe de Moelle et de Thérapie Cellulaire | Bone Marrow Transplantation
Abstract In this SFGM-TC registry study, we report the results after stem cell transplantation (HSCT) in 305 myelofibrosis patients, in order to determine potential risk factors associated with outcomes, especially regarding previous treatment with ruxolitinib. A total of 102 patients were transplanted from an HLA-matched-sibling donor (MSD), and 143 patients received ruxolitinib.
Durable engraftment after pharmacological pre-transplant immune suppression followed by reduced-toxicity myeloablative haploidentical stem cell transplantation in highly HLA-immunized adults with sickle cell disease | Bone Marrow Transplantation
Abstract Allogeneic stem cell transplantation (Allo-SCT) is the only rapidly available curative treatment modality in patients with severe sickle cell disease (SCD). The development of reduced-toxicity myeloablative conditioning (RT-MAC) regimen and the use of partially matched family donors with post-transplantation cyclophosphamide (PT-Cy) have widened the access to Allo-SCT. Antibodies against donor-specific HLA (DSA) increase the risk of engraftment failure in HLA mismatched Allo-SCT.
Allogeneic hematopoietic stem cell transplantation for adults with therapy-related acute myeloid leukaemia: a retrospective multicentre study on behalf of the SFGM-TC | Bone Marrow Transplantation
Abstract We report the results from a multicentre retrospective study of 220 adult patients who underwent allogeneic hematopoietic stem cell transplantation (alloHSCT) for therapy-related acute myeloid leukaemia (t-AML). Median age at t-AML diagnosis was 56 years, with a prior history of haematological (45%) or breast (34%). Median time from cytotoxic exposure to t-AML diagnosis was 54.7 months.
Endowing universal CAR T-cell with immune-evasive properties using TALEN-gene editing - Nature Communications
Abstract Universal CAR T-cell therapies are poised to revolutionize cancer treatment and to improve patient outcomes. However, realizing these advantages in an allogeneic setting requires universal CAR T-cells that can kill target tumor cells, avoid depletion by the host immune system, and proliferate without attacking host tissues.
