Chemotherapy can enhance CAR-cell therapy through several mechanisms. Firstly, it induces lymphodepletion before CAR-T cell infusion. Cyclophosphamide and fludarabine are commonly used to eliminate endogenous immune cells, creating a favourable niche for CAR-T expansion and persistence. Lymphodepletion reduces Tregs and MDSCs, which are known to suppress CAR-T activity.
