Full-length tau protein (referred to as 2N4R), dGAE (PHF-core domain of 2N4R tau from amino acids 297–391) and dGA (tau297-390) proteins were expressed in bacteria and purified as previously described [1]. For sheep immunisation, 2N4R tau and dGAE were used as antigens with a primary inoculum (250 µg) followed by 4 subsequent boosts (125 µg)(Ig Innovations Ltd., Wales, United Kingdom).

Introduction Amongst lysosomal cysteine cathepsins, cathepsin S (CTSS) holds particular interest due to a range of distinctive properties including a normal restricted expression profile, inducible upregulation and activity across a broad pH range (Kirschke et al., 1989; Hsing and Rudensky, 2005; Turk et al., 2012; Wilkinson et al., 2015; Kramer et al., 2017).
