Motivation, Optimism And ... Sleep?
James (Jim) Davis, EdM, MA; National Coach of the Year; Dawson Davis LLC. Driven to perform? You might want to start with your health, since leading well depends on the wellness of the leader. By wellness, I’m not talking about lavender candles and CBD bubble baths; those are nice, but not necessary. What is necessary, as identified by the High Order Performance Framework, is movement, nutrition and rest. Among those foundational components of health, only one of them takes care of itself.
Real-world outcomes with elranatamab in multiple myeloma: a multicenter analysis from the U.S. Multiple Myeloma Immunotherapy Consortium
|
Abstract Elranatamab, a BCMA-CD3 bispecific antibody, has demonstrated robust activity in relapsed/refractory multiple myeloma (RRMM), but real-world outcomes remain poorly defined. We conducted a multicenter retrospective study of 130 patients treated with commercial elranatamab across nine U.S. academic centers. The cohort was heavily pretreated (91% triple-class refractory, 49% penta-refractory), with 49% previously exposed to BCMA-targeted therapies.

Efficacy of stem cell boost (SCB) for chimeric antigen receptor-T cell therapy (CAR-T)-related hematologic toxicity in patients with relapsed/refractory multiple myeloma (RRMM)—real world experience from the US multiple myeloma immunotherapy consortium
|
Abstract Prolonged cytopenias are a well-recognized complication following CAR-T therapy. Autologous SCB offers a potential strategy to promote fast hematologic recovery. We conducted a retrospective multi-institutional comparing outcomes of SCB versus supportive care alone in patients with prolonged cytopenias after CAR-T infusion. Patients were included if they received SCB within 1 year following commercial CAR-T between 6/2021 and 3/2024.

Projections of 21st-century sea-level fall along coastal Greenland - Nature Communications
Abstract The Greenland Ice Sheet will significantly contribute to global mean sea-level rise this century. However, glacial isostatic adjustment is expected to cause regional sea-level fall around Greenland as the land rebounds and the gravitational pull of the shrinking ice sheet decreases.
The impact of extramedullary and paraskeletal plasmacytomas on treatment outcomes in multiple myeloma treated with teclistamab: U.S. Myeloma Immunotherapy Consortium real-world experience | Blood Cancer Journal
|
Abstract Teclistamab, a bispecific antibody targeting B-cell maturation antigen (BCMA), is effective in relapsed or refractory multiple myeloma (RRMM), but its impact on patients with soft tissue plasmacytomas is unclear. We studied 385 RRMM patients treated with teclistamab at 13 U.S. centers through September 2023, with follow-up to April 2024.

Smoking, Obesity, and Post-Cessation Weight Gain: Neurobiological Intersection and Treatment Recommendations
Abstract In the US, 28.8 million adults currently smoke cigarettes, and approximately 1.25 billion people use tobacco globally. Unfortunately, post-cessation weight gain is a substantial barrier to smoking cessation and sustained abstinence. Among people who smoke, 36% meet the body mass index (BMI) criteria for obesity and over 50% meet the waist circumference criteria for central obesity.
Outcomes of elderly patients with relapsed refractory multiple myeloma (RRMM) treated with teclistamab: a multicenter study from the U.S. Multiple Myeloma Immunotherapy Consortium | Blood Cancer Journal
|
Abstract Teclistamab, a BCMA-directed bispecific antibody, received regulatory approval for relapsed/refractory multiple myeloma (RRMM) based on the MajesTEC-1 study. Despite the fact that myeloma is primarily a cancer of elderly adults, only 15% of MajesTEC-1 participants (n = 24) were ≥75 years old. In this multicenter retrospective study, we report real-world outcomes of a large cohort of older RRMM patients treated with teclistamab.

Development of an Endothelial Activation and Stress Index (EASIX)-based predictive model for cytokine release syndrome and neurotoxicity after B-cell maturation antigen directed chimeric antigen receptor T-cell therapy for relapsed/refractory multiple mye
The Endothelial Activation and Stress Index (EASIX) score [lactate dehydrogenase (LDH) (U/L) x creatinine (Cr) (mg/dL)/platelet count (109/L)] is a marker of endothelial dysfunction and was initially developed to predict overall survival (OS) after allogeneic hematopoietic cell transplantation [1].
A real-world experience of efficacy and safety of belantamab mafodotin in relapsed refractory multiple myeloma - Blood Cancer Journal
Abstract While initial trials led to the accelerated approval of belantamab mafodotin, a BCMA-directed antibody-drug conjugate, confirmatory trials failed to establish benefit from this therapy for patients with relapsed refractory multiple myeloma (RRMM), eventually leading to its withdrawal from commercial use. With an imminent approval as an effective combination therapy, as seen in recent randomized trials, we report real-world clinical outcomes with belantamab mafodotin in 81 RRMM patients.
Dexamethasone for the management of CRS Related to teclistamab in patients with relapsed/refractory multiple myeloma - Blood Cancer Journal
TO THE EDITOR: Cytokine release syndrome (CRS) is a systemic inflammatory response triggered by the activation of T-cells and is commonly seen in patients receiving immune effector cell therapies such as chimeric antigen receptor (CAR) T-cell therapy and bispecific antibodies (BsAbs) which target varying tumor antigens such as CD19, CD20, B-cell maturation antigen (BCMA), or GPRC5D depending on the type of malignancy.

Now For Nature: The Decade Of Delivery
This report is co-authored with CDP Companies’ impacts on climate and nature are inextricably linked – and need to be addressed in collaboration with their value chain. The emissions created by companies’ value chains are about six times more than their “direct” operational emissions, and their supply chains reach deep into countries with high rates of deforestation and water stress.
From Ambition To Reality
At COP26 finance took a central role in the global climate agenda for the first time, under the banner of GFANZ. And while industry leaders have already made big strides to build new capabilities, launch new propositions, and to adapt the way they do business, there is far more to do. Corporate banks will be at the centre of driving and helping their clients navigate the transition to a net zero economy.

Biden’s New War on Drugs
By a wide margin, Americans believe the War on Drugs has been a failure and are ready for a different approach. Given that the old drug war has gone so poorly, why does President Joe Biden want to launch a new one, this time on prescription drugs? As senator, Biden was a leading advocate for first enacting and then sustaining the war on illegal drugs. By any objective standard, Biden’s war accomplished few, if any, of its objectives; in fact, the illicit drug market thrived. Read Full Article »
