Editor’s summary Inborn errors of immunity are caused by monogenic mutations that result in dysregulation of the immune system. By performing whole-exome sequencing in patients presenting with recurrent respiratory tract infections, Ransmayr et al. identified that biallelic loss-of-function mutations in the lymphotoxin beta receptor (LTβR) result in an inborn error of immunity characterized by an absence of functional secondary lymphoid organs (SLOs).
