To study the function of FMRP during the early stage of human neural development, we disrupted FMR1 gene in the male iPSC line TIG11428. We confirmed similar expression of the stemness marker stage-specific embryonic antigene-4 (SSEA4) in both the parental (TIG-WT) and a clonal FMR1-KO (TIG-KO) line, and the complete absence of FMR1 mRNA in the TIG-KO (Fig. 1A,B). There was no difference in morphology or growth between these cells.
