1 Introduction The nuclear receptor-binding SET domain protein 2 (NSD2) functions primarily as a histone methyltransferase catalyzing H3K36 dimethylation (H3K36me2) and plays an indispensable role in orchestrating embryonic gene regulation and chromatin architecture (Topchu et al. 2022). Homozygous deletion of Nsd2 in mice leads to late embryonic developmental defects, resulting in reduced neonatal size, respiratory distress, and lethality within 10 days postnatally (Kinoshita et al. 2024).
