Editor’s summary Alternative splicing events may result in protein isoforms that promote tumor progression. Xu et al. found that a cryptic splice site of STAT3 resulted in an isoform of STAT3 lacking S701 (ΔS701), which was abundant in colorectal cancer tissue and associated with increased STAT3 activation. Reversible phosphorylation of S701 could be catalyzed by the actions of mTORC1 and PP2A, and the phosphorylation of S701 restricted the phosphorylation of Y705 and thus reduced STAT3 activation.
