Cryo-EM structures of plant Augmin reveal coiled-coil assembly, antiparallel dimerization, and NEDD1 binding - Nature Communications
Abstract Microtubule (MT) branch nucleation requires Augmin and NEDD1 proteins, which recruit and activate the gamma-tubulin ring complex (γ-TuRC). Augmin is a fork-shaped assembly of eight coiled-coil subunits, while NEDD1 is a β-propeller protein bridging MTs, Augmin, and γ-TuRC. We reconstitute Arabidopsis thaliana Augmin assemblies and determine 3.7-7.3-Å cryo-EM structures of its V-junction and extended regions using crosslinking mass spectrometry.
Teachers & Writers Magazine
Standing amongst the small desks, I nervously await my youngest class ever. Marilyn Kaggen, a special education teacher here at the Patrick Daly School in Brooklyn, must sense my anxiety because she smiles and quotes poet and professor Kenneth Koch: “The power to see the world in a strong, fresh, and beautiful way is a possession of all children. Their degree of literacy does not form their imaginations.” “Right!” her paraprofessional colleague, Joanne Barnes, adds.
Proteins with cognition-associated structural changes in a rat model of aging exhibit reduced refolding capacity
Abstract Cognitive decline during aging represents a major societal burden, causing both personal and economic hardship in an increasingly aging population. Many studies have found that the proteostasis network, which functions to keep proteins properly folded, is impaired with age, suggesting that there may be many proteins that incur structural alterations with age.
Protein misfolding involving entanglements providesa structural explanation for the origin of stretched-exponential refolding kinetics
Abstract Stretched-exponential protein refolding kinetics, first observed decades ago, were attributed to a nonnative ensemble of structures with parallel, non-interconverting folding pathways. However, the structural origin of the large energy barriers preventing interconversion between these folding pathways is unknown. Here, we combine simulations with limited proteolysis (LiP) and cross-linking (XL) mass spectrometry (MS) to study the protein phosphoglycerate kinase (PGK).

The importance of being Uncle Wolf Original
While I had a truly exceptional dad, I have always depended on the kindness of uncles. All kinds of uncles: the ones related by blood, the ones related by marriage, the ones you "call uncle" because they're related by family friendships; the ones you see all the time, the ones you see a few times a year but always on the same specific occasions and ones you see hardly ever, but always wished you saw more.
PROFILES IN MENTAL HEALTH COURAGE by Patrick J Kennedy Stephen Fried Read by Johnny Heller Patrick J Kennedy | Audiobook Review | AudioFile Magazine
by | Read by Johnny Heller, Patrick J. Kennedy After JFK’s nephew, Patrick, delivers the prologue, Johnny Heller’s moving performance of this excellent audiobook shows why he’s regarded as one of today’s top narrators. His varied pacing and subtle voicings and volume modulations make for good listening in themselves. Furthermore, they’re never out of sync with the shifting moods and overall poignant content offered by the articulate congressman and his cowriter.
Cognition-Associated Protein Structural Changes in a Rat Model of Aging are Related to Reduced Refolding Capacity
Abstract Cognitive decline during aging represents a major societal burden, causing both personal and economic hardship in an increasingly aging population. There are a few well-known proteins that can misfold and aggregate in an age-dependent manner, such as amyloid β and α-synuclein.

Abstract Here, we present FLiPPR, or FragPipe LiP (limited proteolysis) Processor, a tool that facilitates the analysis of data from limited proteolysis mass spectrometry (LiP-MS) experiments following primary search and quantification in FragPipe. LiP-MS has emerged as a method that can provide proteome-wide information on protein structure and has been applied to a range of biological and biophysical questions.
Lowering Activation Barriers to Success in Physical Chemistry (LABSIP): A Community Project Original
Physical chemistry is a major pillar of the undergraduate curriculum. In many four-year colleges and universities in the United States, the chemistry major requires two semesters of physical chemistry (and their associated laboratory courses), during which students are often first exposed to the foundational ideas and equations of quantum mechanics, thermodynamics, and kinetics.
Is the secret to a happy marriage learning to be alone?
During the early years of marriage, my primary leisure activity was being married. It was all so new and different. I was still astonished at always having a date. My wife, Diane, was perfectly happy to have an attentive husband but eventually she started to wonder what was wrong with me - especially when the weather would get warm, and regular guys were out doing regular guy things. Of course, I took this to mean that I should find something new for us to do together: tennis, yoga classes, chess.

How synonymous mutations alter enzyme structure and function over long timescales - Nature Chemistry
Abstract The specific activity of enzymes can be altered over long timescales in cells by synonymous mutations that alter a messenger RNA molecule’s sequence but not the encoded protein’s primary structure. How this happens at the molecular level is unknown.
Stephen Fried: Ted Turner’s New Mexico ranch brings back American West

Accurate Protein Domain Structure Annotation with DomainMapper
Abstract Automated domain annotation plays a number of important roles in structural informatics and typically involves searching query sequences against Hidden Markov Model (HMM) profiles. This process can be ambiguous or inaccurate when proteins contain domains with non-contiguous residue ranges, and especially when insertional domains are hosted within them.
Nonrefoldability is Pervasive Across the E. coli Proteome
Note Made available for a limited time for personal research and study only License. Introduction Many experiments of protein folding, conducted on purified, small, single-domain, soluble proteins, follow the proportion of protein molecules that are folded as a function of time, temperature, denaturant concentration, or sequence (1−3) and have yielded immense insight into the molecular determinants that underpin stable globular folds.
The Founders’ Plague-and Ours
The quick development of safe, effective COVID-19 vaccines has been one of the great triumphs in U.S. medical history. Nobody would have been more delighted to see it than the nation’s first great physician, the patriotic Philadelphian Benjamin Rush. A strong believer in inoculations to prevent smallpox, he pressed fellow delegates to the Second Continental Congress to get them.

Remote Work During the Pandemic: What We Got Wrong
By Stephen Fried, CISSP As COVID-19 began to spread rapidly across the globe in 2020, many organizations moved their employees off company premises and enabled large "work from home" efforts. Nobody knew how long this would last, but we assumed we could work remotely for a few months until this thing worked itself out, then return to the office and get back to "normal." We were very wrong.
Securing New Technology Is Going To Be Ok. Just Not Today.
Stephen Fried, IT Security Director, American Family Mutual Insurance Company, S.I. My cell phone has a camera in it. That’s not a big deal today, but back in 2003 cell phone cameras were a cutting-edge technology and well-meaning security pros (myself included) warned that this would allow attackers to steal all your data, violate your privacy rights, and cause the downfall of our companies. Ah, the good old days, when camera phones were our biggest problem.
MOTHER TERESA AND ME
The call came, as I had always known it one day would. "We're making a TV series called J'Accuse, about inflated and bogus reputations. Do you have a nominee?" With my voice catching only slightly, I replied, "Well, I presume someone's already snapped up old Mother Teresa." But, no, the Down with Mother Teresa slot was vacant. At last. At last! Should I have suspected my motives here? Hardly necessary, since other people would do that for me. But still, why was I so sure this job needed doing?
