The final results of a multicenter phase II study of TAS-102 monotherapy in patients with pre-treated advanced gastric cancer (EPOC1201)
Background: American phase I studies have reported that the recommended dose of TAS-102 (trifluridine/tipiracil) was 25 mg/m2 b.i.d., although this schedule did not provide clinically relevant improvements in a phase II study of advanced gastric cancer (AGC). However, a pivotal phase III study revealed that TAS-102 at 35 mg/m2 b.i.d. provided a clinically relevant improvement in overall survival among patients with metastatic colorectal cancer.
Phase I dose escalation (esc) trial of weekly intravenous (i.v.) BI 836845 in Japanese patients (pts) with advanced solid tumors
Background: BI 836845 is an insulin-like growth factor (IGF) ligand-neutralizing humanized antibody that binds to IGF-1/IGF-2 ligands. This Phase I trial (NCT02145741) evaluated the maximum tolerated dose (MTD), safety, pharmacokinetics (PK), pharmacodynamics (PD), and activity of BI 836845. Methods: Japanese pts with advanced tumors were recruited in a 3 + 3 dose esc design. BI 836845 was administered weekly i.v. (21-day cycles).
