An episignature informed systematic analysis to ascertain the clinical significance and consequences of CHD8 missense variants
Abstract Pathogenic CHD8 variants cause autosomal dominant ‘Intellectual developmental disorder with autism and macrocephaly’ (IDDAM) and are amongst the most common monogenic causes of autism. The clinical significance of CHD8 missense variants (MVs) frequently remains uncertain. Systematically applying ACGS/ACMG guidelines to 36 CHD8 MVs in 39 affected patients, only two variants were classified likely pathogenic (LP), with the remaining 34 classified variants of uncertain significance (VUS).
Systematic mapping of rare genetic disease studies using UK primary care electronic health records
Abstract Rare disease studies often rely on small, selected cohorts, are resource-intensive and difficult to scale. UK primary care electronic health record (EHR) databases provide population-based, longitudinal data, but their use for rare genetic disease research has not been systematically examined.

Isolation and Bioassay of Linear Veraguamides from a Marine Cyanobacterium (Okeania sp.)
Abstract Marine cyanobacteria have gained momentum in recent years as a source of novel bioactive small molecules. This paper describes the structure elucidation and pharmacological evaluation of two new, veraguamide O (1) and veraguamide P (2), and one known, veraguamide C(3), analogs isolated from a cyanobacterial collection made in the Las Perlas Archipelago of Panama. We hypothesized that these compounds would be cytotoxic in cancer cell lines.

Clinical validation of p16/Ki‐67 dual‐stained cytology triage of HPV‐positive women: Results from the IMPACT trial
Abstract Triage strategies are needed for primary human papillomavirus (HPV)-based cervical cancer screening to identify women requiring colposcopy/biopsy. We assessed the performance of p16/Ki-67 dual-stained (DS) immunocytochemistry to triage HPV-positive women and compared it to cytology, with or without HPV16/18 genotyping. A prospective observational screening study enrolled 35 263 women aged 25 to 65 years at 32 U.S. sites.
Arthroscopically Assisted Surgical Decompression and Fibular Strut Grafting for Proximal Humerus Avascular Necrosis: Surgical Technique
Abstract Avascular necrosis is a relatively common entity that affects the proximal humerus and can lead to substantial morbidity. It often occurs in younger patients for whom the traditional treatment of shoulder arthroplasty is not optimal. Fibular strut grafting to prevent humeral head collapse has been described as a viable treatment option. However, it is technically challenging to direct the fibular strut graft into the center of the bony infarct, where it will be most effective.
Is data recovery possible on an SSD?
This is a sponsored article by Salvage Data. For a variety of reasons, SSDs can be damaged and fail. If your SSD fails, it can be a great if the data on it is important to you, such as work files or treasured family photos. But luckily, there are some changes to recover SSD data. Plenty of data recovery companies provide SSD data recovery services. But before relaxing, you need to understand that it is much more difficult to recover SSD data than HDD.
Clinical Utility of Extended Genotyping
Date: October 7, 2020 Time: 9:00am (PDT), 12:00pm (EDT) Recently the FDA approved the BD Onclarity™ HPV assay as a test to identify HPV high-risk genotypes in addition to HPV 16 and 18 (i.e., extended genotyping). The Onclarity assay is designed to identify the 14 high-risk HPV genotypes either individually (HPV 16, 18, 31, 45, 51, 52) or as a pooled mixture of two to three genotypes (HPV 33/58, 35/39/68, 56/59/66).

H-Bonding Organocatalysts for Ring-Opening Polymerization at Elevated Temperatures
The ring-opening polymerization (ROP) kinetics of ε-caprolactone and lactide with various H-bonding organocatalysts, (thio)ureas paired with an amine cocatalyst, were evaluated at temperatures up to 110 °C. In nonpolar solvent, most cocatalyst systems exhibit decomposition at high temperatures, while only two, a monourea and bis-urea H-bond donor plus base cocatalyst, are stable up to 110 °C. The onset temperature of cocatalyst decomposition must be measured under reaction conditions.
Azithromycin treatment modifies airway and blood gene expression networks in neutrophilic COPD
Abstract Long-term, low-dose azithromycin reduces exacerbation frequency in chronic obstructive pulmonary disease (COPD), yet the mechanism remains unclear. This study characterised genome-wide gene expression changes in patients with neutrophilic COPD following long-term, low-dose azithromycin treatment. Patients with neutrophilic COPD (>61% or >162×104 cells per mL sputum neutrophils) were randomised to receive either azithromycin or placebo for 12 weeks.
Two Hundred Years of Magma Transport and Storage at Kīlauea Volcano, Hawaiʻi, 1790-2008
This publication summarizes the evolution of the internal plumbing of Kīlauea Volcano on the Island of Hawaiʻi from the first documented eruption in 1790 to the explosive eruption of March 2008 in Halemaʻumaʻu Crater. For the period before the founding of the Hawaiian Volcano Observatory in 1912, we rely on written observations of eruptive activity, earthquake swarms, and periodic draining of magma from the lava lake present in Kīlauea Caldera.
IBP for Retail enterprises, it’s a viable and valuable process
I did a quick post on LinkedIn earlier in the week on application of IBP in a retail business. Subsequent conversations have suggested there is sufficient interest in the topic to capture a few more thoughts here and hopefully start a conversation. Like most conversations about applications of IBP, the ides aren’t new. My friends at Ollie Wight did a great three part overview of Retail IBP back in 2010. And the best run companies we work with of course have a planning process in place.
A little “live” experience with IBP Demand
Presales, my role at SAP, provides a variety of interesting customer engagements, and as I’ve shared earlier on this blog channel, in a best case outcome there is learning for us and for our customers. That’s certainly the case in a recent experience, where I led a team providing a customized demonstration of IBP Demand and Inventory using a customer provided dataset. We don’t always – or even often – do this. There needs to be a win/win outcome to justify the mutual investment.
IBP for MRO - of course it’s all about the Forecast!
I had the chance last night to speak with an IBP customer starting a deployment to manage MRO for a set of mobile and fixed assets. While nothing I say here is confidential, I will respect their privacy as i did not request permission to “name names”. But they are of course free to provide an identifying comment. We talked about a broad range of topics impacting planning for MRO, they are well informed and have been in contact with SAP executives in both IBP and Asset Management domains.
So you’re starting an Integrated Business Planning project - Part 4
“Without data, you’re just a person with an opinion” I learned this quote from Edwards Deming during the quality phase of my professional development. But I’ve also learned it applies to a broad range of activities, and absolutely applies to IBP. In this fourth and last blog in this series, we’ll discuss data. Hold on to your hats, this is exciting! Well, maybe not, but it is critical.
So you’re starting an Integrated Business Planning project - Part 3
“When you give a five year old a hammer, the world suddenly becomes filled with things that need to be hammered” I think credit for this goes to Abraham Kaplan…I love that quote. It’s one I’ve used many times in many situations, but perhaps nowhere does it fit better than when discussing scope management (aka scope creep) associated with an IBP project. That’s the topic of the third blog in this series. In part one, we talked about change management.
So you’re starting an Integrated Business Planning project - Part 2
In our last conversation we discussed the imperative of change management, without a program to ensure this is done effectively there is no point in getting started. But there is more than just managing change. As they say, a key prerequisite from change is knowing what to change to. That’s the topic of this bog, part 2 in this series. In “The Seven Habits of Highly Effective People”, Steven Covey includes “Begin with the end in mind”.
So you’re starting an Integrated Business Planning project - Part 1
I’m just leaving our 2017 Kickoff meeting and talked with many colleagues and partners who have customers launching IBP projects shortly. So it seems like a great time to expand on content in my book ( https://www.sap-press.com/sap-integrated-business-planning-how-does-sap-ibp-fit-into-scm-processes_4174/ ) with a little more detail on topics to keep in mind as your team starts this important journey.
Consuming IBP-Generated PIR’s in ERP
How’s that for three TLA’s in one title? But it’s an important topic that I discuss often when describing how IBP will integrate back with ERP to operationalize an approved plan.
Managing ERP Inventory Planning with IBP Inventory
Over the holiday slowdown I had the chance to do some deep-dive testing in the new IBP for inventory capabilities in the 1608 and beyond releases, particularly the ability to calculate additional inventory components ( ie, cycle stock, pipeline stock) based on forecast demand and knows safety stock.
Introduction to Lipedema
Although lipedema was first described by Drs. Edgar Allen and Edgar Hines in 1940 at the Mayo Clinic, many physicians remain unaware of this medical condition or its symptoms (Hines, 1940). Lipedema is a chronic, progressive medical disease that causes a disproportionate amount of swollen fat deposition in the arms and/or legs. As lipedema worsens, the affected limbs feel heavy and can be painful. The condition is associated with easy bruising, especially in the affected limbs.
Map Showing Lava-Flow Hazard Zones, Island of Hawaii
Discussion This map shows lava-flow hazard zones for the five volcanoes on the Island of Hawaii. Volcano boundaries are shown as heavy, dark bands, reflecting the overlapping of lava flows from adjacent volcanoes along their common boundary. Hazard-zone boundaries are drawn as double lines because of the geologic uncertainty in their placement. Most boundaries are gradational, and the change In the degree of hazard can be found over a distance of a mile or more.
