Therapeutic activation of farnesoid X receptor (FXR) has long been pursued as a way to correct bile acid dysregulation in chronic liver diseases, but high affinity long-acting FXR agonists have often faced a narrow therapeutic window. Recently, Zang, Shi, Zhao et al. reported that linafexor (CS0159) is a first-in-class, non-bile-acid FXR agonist designed for rapid clearance and transient receptor activation [1].
