Abstract DNA methylation patterns at crucial short sequence features, such as enhancers and promoters, may convey key information about cell lineage and state. The need for high-resolution single-cell DNA methylation profiling has therefore become increasingly apparent. Existing single-cell whole-genome bisulfite sequencing (scWGBS) studies have both methodological and analytical shortcomings.

Abstract DNA methylation is a relatively stable epigenetic mark with important roles in development and disease. Since cell-to-cell variation in epigenetic programming can reflect important differences in cell state and fate, it is clear that single-cell methods are essential to understanding this key epigenetic mark in heterogeneous tissues.

Abstract Imprinting disorders (IDs) often affect growth in humans, leading to diseases with overlapping features, regardless of the genomic region affected. IDs related to hypomethylation of the human 14q32.2 region and its DLK1/MEG3 domain are associated with Temple syndrome (TS14). TS14 is a rare type of growth retardation, the clinical signs of which overlap considerably with those of Silver-Russell syndrome (SRS), another ID related to IGF2 down-regulation at 11p15.5 region.
