Baseline gut microbiome features associated with fecal calprotectin response to exclusive enteral nutrition in pediatric Crohn’s disease
Abstract Background and aims: Exclusive enteral nutrition is an established induction therapy for pediatric Crohn's disease, but biochemical response varies across patients. We assessed whether baseline gut microbiome features were associated with fecal calprotectin response and whether treatment-related taxonomic shifts could be interpreted alongside reported fecal biochemical summaries. Methods: We re-analyzed a public pediatric cohort (PRJEB14084).
A reproducible pretreatment mucosal-inflammatory-remodeling state is associated with induction-phase anti-tumor necrosis factor non-response in ulcerative colitis
Abstract Background: Primary non-response to anti-tumor necrosis factor (anti-TNF) therapy remains a major clinical challenge in ulcerative colitis (UC); however, public-data transcriptomic studies often yield unstable single-gene biomarkers and limited replication. Methods: We analyzed three independent pretreatment UC mucosal transcriptome cohorts from infliximab-treated patients (total n = 79). The induction response was assessed at 4–8 weeks according to the original definition in each study.

Ppid is Necessary for Overnutrition-Induced β-cell Loss
Type 2 diabetes (T2D) involves progressive loss of functional β-cell mass. In a zebrafish insulin-resistant model (zMIR), overnutrition triggers islet inflammation and nocturnal β-cell death. The cell death is prevented by the cyclophilin D (Ppid) inhibitor, cyclosporin A (CsA). Reducing mitochondrial ROS with mito-TEMPO or mitochondrial calcium with Ru360 protects β cells, further implicating the mitochondrial permeability transition pore (mPTP) in β-cell loss.
Hepatitis B virus X protein promotes the progression and immune escape of hepatocellular carcinoma by activating KLF16-C12orf49-PD-L1 axis - Oncogene
Abstract Hepatocellular carcinoma (HCC) is a common and serious type of malignant tumor with an unfavorable prognosis, partly attributed to the prevalence of hepatitis B virus (HBV) infection. However, the molecular mechanism underlying HBV-HCC are not yet fully understood. Here, we found that Kruppel-like factor 16 (KLF16) was significantly upregulated in HBV-HCC and KLF16 knockdown suppressed the growth and metastasis of HBV-infected HCC cells.

Plaat1l1 controls feeding induced NAPE biosynthesis and contributes to energy balance regulation in zebrafish.
Abstract Dysregulation of energy balance leading to obesity is a significant risk factor for cardiometabolic diseases such as diabetes, non-alcoholic fatty liver disease and atherosclerosis. In rodents and a number of other vertebrates, feeding has been shown to induce a rapid rise in the intestinal levels of N-acyl-ethanolamines (NAEs) and the chronic consumption of a high fat diet abolishes this rise.

Fact or Artifact in Inferior ST Elevation
Challenges in Clinical Electrocardiography Fact or Artifact in Inferior ST Elevation—Avoiding Misdiagnosis and Missed Diagnosis A patient in their 60s with a history of hypertension presented to the emergency department with sudden chest pain radiating to the back and sweating for 2 hours without relief. Initial physical examination results revealed blood pressure of 140/78 mm Hg, a pulse rate of 74/min, and a respiratory rate of 20/min.

Shark Fin Electrocardiogram in the Intensive Care Unit
Originally publishedhttps://doi.org/10.1161/CIRCULATIONAHA.122.062034Circulation. 2022;146:1099–1102 ECG Challenge A 54-year-old woman was admitted to the intensive care unit with septic shock induced by intraabdominal infection. An initial physical examination revealed a body temperature of 38.6°C, a blood pressure of 82/56 mm Hg, a pulse rate of 109 beats per minute, and a respiratory rate of 26 breaths/minute. At admission, a 12-lead ECG was performed (Figure 1).
A Vegfc-Emilin2a-Cxcl8a Signaling Axis Required for Zebrafish Cardiac Regeneration
Originally publishedhttps://doi.org/10.1161/CIRCRESAHA.121.319929Circulation Research. 2022;130:1014–1029 Other version(s) of this article Abstract Background: Ischemic heart disease following the obstruction of coronary vessels leads to the death of cardiac tissue and the formation of a fibrotic scar. In contrast to adult mammals, zebrafish can regenerate their heart after injury, enabling the study of the underlying mechanisms.

Microinjection of CRISPR/Cas9 Protein into Channel Catfish, Ictalurus punctatus, Embryos for Gene Editing
Microinjection is a common laboratory technique used to deliver a small amount of a substance such as DNA, RNA, protein, and other macromolecules into cells or embryos through a glass capillary1. Microinjection is performed using special equipment setup including a microinjector, micromanipulator, and a microscope2.
