Transforming growth factor-β (TGF-β) is a potent immunosuppressive cytokine abundant in the tumor microenvironment. As an intracellular antagonist of TGF-β signaling, SMAD7 counteracts TGF-β-mediated immunosuppression and thereby plays an important role in regulating antitumor immune responses. However, the specific functions of SMAD7 in natural killer (NK) cells, which are critical effectors in antitumor immunity, remain poorly defined.
