1 Introduction DesII is a radical S-adenosyl-l-methione (SAM) enzyme that catalyzes the radical-mediated deamination of TDP-4-amino-4,6-dideoxy-d-glucose (1) to TDP-3-keto-4,6-dideoxy-d-glucose (2) during the biosynthesis of TDP-desosamine (3, Figure 1A).[1-4] Like other radical SAM enzymes,[5, 6] the catalytic cycle of DesII is characterized by initial single electron transfer from a reduced, active site [Fe4S4]+ cluster to SAM (Figure 1B).
