Macrophage pyroptosis driven by Omicron variant’s distinct entry route underlies severe disease pathogenesis
Abstract Severe disease due to SARS-CoV-2 infection is a primary driver of healthcare burden throughout the pandemic. Although the overall pathogenicity of the Omicron variant is attenuated compared with earlier strains, mortality among patients who develop severe disease following Omicron infection remains substantial, and the underlying mechanisms remain unclear.
Low-dose oral nicotinamide mononucleotide for immune thrombocytopenia: a phase 1/2 trial
Abstract Autoimmunity remains challenging to treat without broad immunosuppression. We previously showed that anti-CD38 antibody can rapidly elevate platelet counts in refractory immune thrombocytopenia (ITP), but the underlying mechanism was unclear. Here we report that anti-CD38 antibody induces platelet recovery within 3 days, including after retreatment in relapsed cases.
Variant-divergent death: Omicron intensifies bystander T-cell apoptosis via GDF15–BCL2L13
Abstract Severe Omicron cases present profound lymphocytopenia, suggesting variant-specific immune injury. We identify CD63 as a conserved T-cell host factor supporting ACE2-independent SARS-CoV-2 entry. Despite lower intracellular viral loads than the ancestral strain, Omicron elicits enhanced T-cell apoptosis largely through a bystander mechanism.
Phase 1 pilot study for hemophilia-A: AAV8 vector with prophylactic tacrolimus-glucocorticoid achieves therapeutic FVIII activity
Abstract This prospective phase 1 pilot study evaluated the safety and efficacy of GS001, an adeno-associated virus serotype 8 (AAV8)-based vector encoding B-domain–deleted factor VIII with exploratory assessment of prophylactic immunosuppression. Twelve adult severe hemophilia A participants were enrolled.

Childhood and adolescent essential thrombocythemia and prefibrotic primary myelofibrosis: insights into diagnosis, outcomes, and treatment from a large Chinese cohort | Leukemia
Abstract The paucity of essential thrombocythemia (ET) and prefibrotic primary myelofibrosis (pre-PMF) in individuals younger than 18 years highlights several unresolved issues in diagnosis, clinical outcomes, and treatment strategies. To address these knowledge gaps, we analyzed a large bidirectional cohort consisting of childhood and adolescent ET (CAA-ET, n = 156) and pre-PMF (CAA-preMF, n = 13), as well as adult ET (n = 349).
Reconstruction of the cell pseudo-space from single-cell RNA sequencing data with scSpace - Nature Communications
Abstract Tissues are highly complicated with spatial heterogeneity in gene expression. However, the cutting-edge single-cell RNA-seq technology eliminates the spatial information of individual cells, which contributes to the characterization of cell identities.

High‐efficient generation of VCAM‐1+ mesenchymal stem cells with multidimensional superiorities in signatures and efficacy on aplastic anaemia mice
Abstract Objective Longitudinal studies have indicated VCAM‐1+ mesenchymal stem/stromal cells (MSCs) as promising resources in regenerative medicine, yet the abundance in gene expression is far from adequate in the advantaged and “discarded” hUC‐MSCs. Thus, high‐efficient preparation and systematic dissection of the signatures and biofunctions of the subpopulation is the prerequisite for large‐scale clinical applications.
The regulation of regulation: IL-10 increases CD4+CD25+ regulatory T cells but impairs their immunosuppressive activity in murine models with schistosomiasis japonica or asthma - He - 2017 - Immunology - Wiley Online Library
CD4+CD25+Foxp3+ regulatory T cells (Tregs) play an important role in maintaining immune homeostasis. Interleukin 10 (IL-10), a cytokine with anti-inflammatory capacities, also has a critical role in controlling immune responses. In addition, it is well known that production of IL-10 is one of suppression mechanisms by Tregs. However, the action of IL-10 on Tregs themselves remains insufficiently understood. In this study, by using a Schistosoma japonicum (S.
A canonical role of SMAD4 in safeguarding 3D genome architecture to suppress lung tumorigenesis
|
Abstract Dysregulation of three-dimensional (3D) genome architecture is a hallmark of cancer, yet the mechanisms by which its disruption drives tumorigenesis remain incompletely understood. Here, we demonstrated that SMAD4 regulated 3D genome organization through its canonical transcription factor activity by directly binding chromatin, thereby suppressing lung tumorigenesis. Hi-C analyses of human lung tumors identify SMAD4 as a potential regulator of 3D genome integrity.
