ERK1/2 function as a central hub safeguarding primed human embryonic stem cell identity via multifaceted regulatory networks
Results To systematically define the function of ERK1 and ERK2 in hESCs, we utilized stepwise CRISPR-Cas9n genetic editing methods to deplete ERK1 and ERK2 in hESCs (Figure 1A and S1A). Briefly, CRE hESCs constitutively expressing ERT2CREERT2 served as control cells. The inducible KO (iKO) of ERK1 or ERK2 was achieved by 4-hydroxytamoxifen (4OHT)-mediated recombination of LoxP sites. An ERK1/2 iKO line (ERK1Δ/Δ; ERK2fl/-) was generated from the ERK1 KO line.
