Low-dose baricitinib plus danazol in primary immune thrombocytopenia: a randomized, controlled phase 2 trial
Abstract This multicenter, randomized, controlled study was conducted to explore the safety and efficacy of low-dose baricitinib plus danazol for ITP patients who had failed corticosteroids and at least one recommended subsequent treatment. Participants were randomly assigned to receive baricitinib plus danazol (n = 108) or danazol alone (n = 108) by a central, interactive web-based system. Patients and caregivers were not blinded to group assignment.

Presently, haploidentical stem cell transplantation (haplo-SCT) has emerged as an alternative therapy for patients with myeloid malignancies [1]. Despite remarkable advances in haplo-SCT, relapse remains the primary cause of treatment failure [2]. Notably, second SCT (SCT2) is the main therapy for patients who relapse after the first haplo-SCT, with 2-year overall survival (OS) of 28.7% [3]. However, the optimal donor for SCT2 is still debated [4,5,6].

To the Editor: Allogeneic hematopoietic stem cell transplantation (allo-HCT) is considered a curative treatment modality for Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) and has improved the long-term survival since the integration of tyrosine kinase inhibitors (TKIs) into chemotherapy and post-transplant maintenance [1]. However, relapse still occurs in up to 30% of patients; in that case, survival outcomes are still unsatisfactory [2].
