Results and discussion To elucidate the CES1/CES2 enzymatic cascade conversion and HSA-binding properties, five CPT prodrugs were systematically synthesized through esterification with aliphatic alcohols of different tail lengths (C6-C14) using succinic acid as a linker. The synthetic routes of prodrugs 1a-e and CPT-SA 4 are detailed in Scheme 1.
