Intratumoral T reg cell ablation elicits NK cell–mediated control of CD8 T cell–resistant tumors
Editor’s summary Loss of MHC class I (MHC I) expression is a key strategy used by cancer cells to evade detection by the immune system. Natural killer (NK) cells can target MHC I–deficient cancers, but their antitumor responses are typically constrained because of other immunosuppressive factors like the presence of regulatory T cells (Treg cells). The rise of spatial TCR profiling: Emerging technologies and open challenges
Abstract High-definition spatial TCR profiling illuminates how clonal identity and T cell state are coordinated in the tumor microenvironment (see related Research Article by McCord et al.). Single-cell TCR mapping reveals spatially coordinated T cell states in head and neck cancer
Editor’s summary Spatial transcriptional profiling of individual T cell clones can provide insight into how T cells function and differentiate in the tumor microenvironment (TME), but technical limitations have made analysis at true single-cell resolution challenging. Guided by single-cell RNA and T cell receptor (TCR) sequencing, McCord et al. designed patient-specific TCR probes to spatially map gene expression in individual T cell clones in human head and neck tumors.
