2. Materials and Methods 2.1. Extraction and Detection of dsRNA Strain BS-5, belonging to R. solani AG-3 TB, was isolated from tobacco leaves. Strain BS-5 was cultured on potato dextrose agar plates for two days. Then, we collected the mycelia and transferred them to potato dextrose broth at 28 °C with orbital shaking at 100 rpm for one week for the subsequent extraction of double-stranded RNA (dsRNA) and total RNA [34,35].
