Transplantation of active mitochondria condensed in liquid–liquid phase-separated hydrogels ameliorates myocardial ischemia-reperfusion injury
Abstract Mitochondrial dysfunction is a major contributor to myocardial ischemia-reperfusion injury, and limits cardiac recovery after blood flow is restored. Although mitochondria transplantation may help restore cellular energy metabolism, its therapeutic benefit is reduced by extracellular calcium-induced mitochondrial damage. Here we show that a thermosensitive phase-separated hydrogel made of gelatin and PEG can condense, protect and deliver freshly isolated mitochondria.
Cardiac reprogramming via transient overexpression of P-glycoprotein alleviates doxorubicin-induced cardiotoxicity in mice and pigs
Abstract Doxorubicin-induced cardiotoxicity represents a significant clinical adverse effect associated with cancer chemotherapy treatment. Inspired by cancer drug resistance mechanisms, we propose a novel strategy termed transient overexpression of p-glycoprotein for cardiac reprogramming to induce cardiac drug resistance as a treatment for cardiotoxicity.
Cardiac Reprogramming with Drug Resistance Alleviates Doxorubicin-induced Cardiotoxicity in Mice and Pigs
Abstract Doxorubicin (Dox)-induced cardiotoxicity represents a significant clinical adverse effect associated with cancer chemotherapy treatment. The systematic toxicity associated with the currently available drug dexrazoxane has limited its clinical utility. Inspired by cancer drug resistance mechanisms, we propose a novel strategy termed Transient overexpression of p-glycoprotein (P-gp) for Cardiac reprogramming (TopCare) to induce cardiac drug resistance as a treatment for cardiotoxicity.
